Abstract:
The purpose of this presentation is to describe and demonstrate two techniques of phlebotomy, the Micromethod of obtaining a hematocrit, the preparation of peripheral blood films, and the operation of the Coulter Counter Model S to determine seven blood parameters. This objective is achieved with the aid of an adult female clinical subject, laboratory technicians, diagrams, and histologic slides. In this presentation the technique of phlebotomy is first demonstrated step-by-step using the syringe technique. The Vacutainer technique is similarly demonstrated. The Microhematocrit method of hematocrit determination is then demonstrated step-by-step. In this method capillary tubes are filled approximately 3/4 full with blood and centrifuged for four minutes and placed on the Microhematocrit reader. The reading for the hematocrit is then taken directly from the reader. Blood films are prepared by placing a small drop of the blood sample approximately 1/2 inch from the end of a laboratory slide. A second slide is used to produce a thin edge of film across the bottom slide. The procedure is repeated for emphasis and a properly stained slide is presented to demonstrate its appearance. Theuse of the Coulter Counter Model S is then demonstrated to determine by means of electronics and pneumatics the white blood count, red blood count, hemoglobin, hematocrit, mean cell volume, mean cell hemoglobin, and mean cell hemoglobin concentration. The program notes that this machine can also be used for prediluted blood as well as for whole blood.
