Dear Dan, As you know by now, I have written to Ching-Juh about our progress. I had not answered sooner because I was feeling quite tentative about the purification and did not want to predict success. As you can see from the gel photo, the mutant was a fairly small portion of the DNA. Also, the DNA was in excellent condition being quite clear of Form II and cellular DNA. I wanted to do two successive purifications as you suggested. This left me with too little DNA to purify after partial digestion. Therefore I have set up pretty good conditions for making Form III and I will transfect all of the DNA and then select out the full length cloned pieces. We have now gotten a super safe vector as I mentioned to Ching-Juh. Therefore [END PAGE ONE] [BEGIN PAGE TWO] we should be able to transfect early in the year. I will be sure to let you know as soon as we have some clones. Although I have started doing some culture work, I am not entirely satisfied yet. I will work on that harder soon.
